چکیده

The purpose of this investigation was to obtain a com parative measure of DMA repair synthesis in hormonedependent normal mammary tissue and hormone-inde pendent D hyperplastic alveolar nodule (HAN) outgrowth. Treatment of mammary fragments in culture with 5 mm hydroxyurea inhibits 98% of the semiconservative DNA synthesis. Treatment of mammary fragments with methyl methanesulfonate in the presence of hydroxyurea results in a 4to 7-fold higher incorporation of [3H]deoxythymidine into the mammary cell DNA than does treatment with hydroxyurea alone. This hydroxyurea-resistant alkylating agent-induced [3H]deoxythymidine incorporation was studied by CsCI density gradient centrifugation and has been found to represent DNA repair replication. Similar levels of repair replication were found in both normal and preneoplastic D -HAN outgrowth. Autoradiographic anal ysis of mammary fragments and D -HAN outgrowth treated with methyl methanesulfonate plus hydroxyurea revealed that 30 to 50% of the epithelial cell nuclei were lightly labeled. No detectable repair synthesis was found in fat or stromal cells. The average number of grains per labeled nuclei was the same for both expiants. These results suggest that a reduced DNA repair capacity is not associated with the increased sensitivity of D -HAN out growth to the tumorigenic effect of chemical carcinogens.

Effects of low dose radiation on the expression of proteins related to DNA repair requiring Caveolin-1 in human mammary epithelial cells

Background: Radiotherapy is an effective and important therapeutic method for breast cancer, but at the same time it has a radiation-induced bystander effect on normal tissue around the tumor. Repair of double-strand breaks (DSBs) by normal cells can reduce the extent of damage caused by this effect. Caveolin-1 (Cav-1) is an important regulatory molecule in cell signal transduction. However, th...

Growth of Preneoplastic Mammary Epithelial Cells in Serum-free Medium1

The conditions for growing preneoplastic mammary epithelial cells in primary cell cultures with serum-free factor-defined medium were established in a series of experiments. The cells, grown in Dulbecco's modified Eagle's medium with 0.6 mw Ca2+, epidermal growth factor, transferrin, insulin, and Na2SeO3, were fibroblast free, exhibited saturation densities similar to those of cells grown in se...

X-ray microanalysis of electrolyte content of normal, preneoplastic, and neoplastic mouse mammary tissue.

Intracellular sodium, chlorine, and potassium concentrations (mmol/kg dry weight) were determined by electron probe X-ray microanalysis of individual epithelial cells in freeze-dried 2-micrometer sections of mouse mammary tissue which were cut at -30 degrees. A model system was utilized in order to compare elemental content of cells from normal pregnant mammary tissue and preneoplastic and neop...

Phenobarbital, Hexachlorocyclohexane, Steroid Compounds, Liver following Treatment with the Tumor Promoters Enhanced Proliferation of Putative Preneoplastic Cells in Rat

Putative preneoplastic islands were induced in rat liver by diethylnitrosamine or nitrosomorpholine administered either as single high doses or continuously for 40 days at low dose levels. Following recovery periods of 3 weeks to 11 months, islands were identified by means of a positive y-glutamyl transferase reaction and/or altered morphology. DMA synthesis, by means of [3H]thymidine autoradio...

Distinction between the Preneoplastic and Neoplastic State of Murine Mammary Glands by spin-echo NMR

We have, using spin-echo nuclear magnetic resonance spectroscopy, measured the relaxation times and diffusion coefficient of water protons in primary mammary adenocarcinomas of mice. In our biological model, three morphological stages were defined: (a) mammary gland tissue from pregnant mice, (b) preneoplastic nodules, and (c) neoplastic tissue. It was found that neoplastic tissues could be dis...
